The problem
Intervention: Rotavirus vaccination (Rotarix RV1 or RotaTeq RV5) with full vaccination requiring all recommended doses
Who it was tested with: Children aged 6 weeks to 5 years in Slovenia, birth cohorts 2019-2023 (n=89,994); 4% partially vaccinated, 25% fully vaccinated.
Children or communities are compared with a group that did not receive the intervention, or followed over years. This is the design that best separates what the intervention did from everything else that changed.
"We linked Slovenian population registry with vaccination, surveillance, and hospitalization databases for 2019-2023 birth cohorts... We estimated VE using quasi-Poisson regression, adjusting for age, healthcare access, socioeconomic status, and calendar time, with person-time at risk included as an offset." The study used a large linked database with statistical adjustment for known confounders to estimate vaccine effectiveness in a defined population over time.
Observational design limits causal inference; low vaccination coverage in the study population; potential unmeasured confounding despite adjustment for multiple factors
