The problem
Intervention: mRNA COVID-19 vaccine (BNT162b2/Comirnaty) administered according to standard schedule
Who it was tested with: 21 SARS-CoV-2-naïve children with diverse inborn errors of immunity.
The same group is measured before the intervention and again afterwards, with a baseline and an endline and sometimes a midline. Change is visible, but other causes cannot be ruled out.
"Twenty-one SARS-CoV-2-naïve participants with diverse inborn errors of immunity were stratified as high or low responders to the BNT162b2 vaccine based on anti-SARS-CoV-2 spike IgG titers at day 28 post-vaccination." The study measured immune characteristics before vaccination and compared them with post-vaccination antibody responses, identifying baseline biomarkers associated with differential vaccine response in children with inborn errors of immunity.
Small sample size (21 participants); cross-sectional baseline design without longitudinal follow-up; single vaccine type and single timepoint for antibody measurement; no validation cohort; causation between gene expression patterns and vaccine response cannot be established
