The problem
Intervention: Peanut oral immunotherapy with low-dose initiation and slow up-dosing, with longitudinal monitoring of basophil activation and sIgE/sIgG4 as possible biomarkers
Who it was tested with: 32 peanut-allergic children: 16 with high Ara h 2-sIgE (≥40 kU/L) and 16 with lower Ara h 2-sIgE.
The same group is measured before the intervention and again afterwards, with a baseline and an endline and sometimes a midline. Change is visible, but other causes cannot be ruled out.
"Thirty-two peanut-allergic children underwent pOIT, including 16 with high and 16 with lower Ara h 2-sIgE." All children received the treatment and were followed over time with no untreated comparison group, so this is a pre-post design.
Small sample, no comparison group without treatment, and exploratory biomarker analyses that need prospective validation. Results are not shown to apply beyond this clinical setting.
