The problem
Intervention: Epicutaneous immunotherapy using VIASKIN peanut patch (VP250) containing 250 µg peanut protein, applied for 12 months
Who it was tested with: Children aged 1 to 3 years with peanut allergy, including subgroups with atopic dermatitis, asthma, and concomitant food allergies.
Children or communities are compared with a group that did not receive the intervention, or followed over years. This is the design that best separates what the intervention did from everything else that changed.
"EPITOPE was a phase 3, double-blind, placebo-controlled trial designed to assess treatment response to VP250...This subgroup analysis assessed response rates for prespecified subgroups, including children with asthma, AD/eczema, and CFA." The randomised controlled design with placebo comparison and prespecified subgroup analysis provides strong evidence for the intervention's efficacy and safety profile across different child populations with allergic comorbidities.
This is a subgroup analysis with potentially reduced statistical power for individual subgroups. The mechanism underlying higher anaphylaxis rates in certain subgroups receiving active treatment versus placebo is not fully explained. Study limited to children aged 1-3 years.
