The problem
Intervention: BNT162b2 mRNA COVID-19 vaccine given as two doses 3-4 weeks apart to children with paediatric rheumatic diseases on ongoing immunomodulatory treatments
Who it was tested with: 41 children with paediatric rheumatic diseases: 34.1% on conventional DMARDs, 39% on biologic DMARDs, 26.8% on combined therapy.
The same group is measured before the intervention and again afterwards, with a baseline and an endline and sometimes a midline. Change is visible, but other causes cannot be ruled out.
"Forty-one children with paediatric rheumatic diseases were included and were vaccinated with the BNT162b2 mRNA vaccine (two doses of 30 µg administered 3-4 weeks apart). To assess the humoral response, IgG antibodies developed against the S1/Receptor-binding domain (RBD) of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein at baseline and 3-4 weeks after the second dose were measured." The study compares antibody responses within the cohort before and after vaccination to assess whether immunocompromised children can still mount an effective immune response.
Single-cohort design without healthy control group; no measurement of cellular immunity or long-term protection; no assessment of breakthrough infections; relatively small sample size
